How to Reconstitute Peptides for Research: A Laboratory Guide

A step-by-step guide to properly reconstituting lyophilized research peptides using bacteriostatic water — covering dilution math, sterile technique, and storage best practices.

Lyophilized (freeze-dried) peptides must be reconstituted before use in any in vitro or in vivo research application. Proper reconstitution preserves peptide integrity, ensures accurate dosing, and extends solution stability. This guide covers the standard laboratory procedure.

Materials Required

You will need: the lyophilized peptide vial, bacteriostatic water (BAC water) or sterile water for injection, a 1mL insulin syringe, alcohol swabs, and a clean laminar flow environment or still-air box if available. Bacteriostatic water (containing 0.9% benzyl alcohol) is preferred for multi-draw vials as it inhibits microbial growth over 28 days.

Step 1: Calculate Your Dilution

Determine the concentration you need. For a 5mg vial where you want 500mcg/0.1mL (a common research concentration), add 1mL of BAC water to yield a 5mg/mL solution. Use our Reconstitution Calculator to generate precise dilution tables for your specific vial size and desired concentration.

Step 2: Reconstitution Technique

Wipe the rubber stopper of both the peptide vial and BAC water vial with alcohol swabs. Draw the calculated volume of BAC water into the syringe. Insert the needle into the peptide vial at an angle, allowing the liquid to run down the inside wall of the glass — do not inject directly onto the lyophilized cake, as this can degrade peptide structure. Gently swirl (never shake) until fully dissolved. The solution should be clear.

Storage After Reconstitution

Reconstituted peptides should be stored at 4°C (standard refrigerator) and used within 28–30 days. Avoid repeated freeze-thaw cycles. Lyophilized (unreconstituted) peptides are stable at −20°C for 24+ months when stored in a sealed, moisture-free environment.
